چکیده

Three different types of beta-carotene mutants of Phycomyces have been studied. In 2 mutants (Type I) beta-carotene is still the principal carotene but scaled down or up relative to wild type. The carotene mixture of 2 mutants (Type II) consists mainly of phytoene and phytofluene. In Type III (2 mutants) beta-carotene is replaced by lycopene. The examination of the mutants reveals that the receptor pigment is very likely neither beta-carotene nor retinal. Transmission spectra through the growing zone of live sporangiophores of 1 of these mutants which contains less than one-thousandth of the beta-carotene content of wild type show that the receptor pigment extinction is less than 0.003 at its maximum.

Chemical alteration of carotene biosynthesis in Phycomyces blakesleeanus and mutants.

The effects of diphenylamine, dimethyl sulfoxide, streptomycin, AMO-1618, and beta-ionone on the carotene composition of a wild-type and three mutant strains of Phycomyces blakesleeanus have been examined. Diphenylamine increased the phytoene and phytofluene concentrations of all strains while reducing the levels of the color carotenes. Dimethyl sulfoxide reduced the concentration of both cycli...

Carotenogenic enzymes from phycomyces

Cell extracts from car mutants of Phycomyces blakesleeanus, which can convert '4C—labelled mevalonic acid into unsaturated carotenes, were treated with high ionic strength buffer and a range of detergents in an attempt to solubilise the carotenogenic enzymes. Phytoene synthetase was dislodged from the membrane with high molarity Tris-HC1 buffer, but the enzymes which convert phytoene into s—car...

Metabolite profiling of Phycomyces blakesleeanus carotene mutants carS

The fungus Phycomyces blakesleeanus provide a renewable biosource of industrial high value compounds such as carotenes, other isoprenoids, organic acids and fatty acids. Several Phycomyces mutants involve in the β-carotene biosynthetic pathway have been isolated. The C5 mutant produces phytoene, due to a null mutation in the carB gene encoding the phytoene dehydrogenase. The S324 mutant accumul...

Inhibition of Carotene Biosynthesis in Cell Extracts of Phycomyces blakesleeanus

Cell extracts o f the Cl 15 (^-carotene-accumulating) strain o f Phycomyces blakesleeanus were incubated with either [2-l4C]MVA or [1-I4C]IPP and a range o f possible inhibitors of carotenogenesis, including bleaching herbicides, biphenyl com pounds and geranylacetone. Several o f these compounds were potent inhibitors o f /7-carotene formation and caused the accumulation o f phytoene. No other...

Evidence on intracellular sites of terminal synthesis of carotenoids in Mucor hiemalis Wehm.

Introduction Available evidence on carotenoid biosynthesis indicates that a general pathway is utilized in all carotenogenic systems, at least in the primary stages (Jensen 1965). The sequential dehydrogenation of the colorless Co precurs~rs to the colored carotenes has been extensively studied in the photosynthe ... t":'"ic----........bacteria (Stanier 1959) and appears to follow the Porter-Li...
